Check nitrite as a separate water parameter
The Colombo NO2 kit measures nitrite using one liquid reagent and a five-minute colour-development period. It is the individual nitrite test in the Colombo range, manufacturer reference 05020287 and SKS code TK9544. The manufacturer’s current product information specifies material for 40 nitrite tests.
Nitrite is produced during the biological processing of ammonia. Colombo identifies a zero nitrite reading as the desired result and explains that a new biological filter needs time to develop its bacterial population. Testing this parameter gives you an actual reading to record when checking a pond or establishing a filter.
Nitrite is not nitrate
NO2 and NO3 look similar on a shopping list, but they identify different tests. This NO2 kit measures nitrite. The Colombo NO3 nitrate kit measures nitrate and has a different reagent sequence, sample volume and development time. Check the chemical abbreviation as well as the product name when ordering a replacement.
The distinction also matters when interpreting results: a nitrate result does not establish that nitrite is zero. Keep separate columns for the two parameters in your records so you can follow each measurement independently rather than combining them under one general nitrogen reading.
Manufacturer test method

This is a single-reagent method. The published nitrite instructions do not include the powder used in Colombo’s nitrate test or the three liquids used for ammonia. Read the bottle label before dispensing it, particularly when several different kits are open on the same work surface.
Follow any revised directions provided with your actual kit. Have a timer ready so the five-minute stage is measured consistently. Comparing the colour immediately after shaking does not complete the published procedure, and leaving an unlabelled tube aside makes it easier to lose track of its development time.
Make the colour comparison in the intended way
Colombo’s leaflet directs you to stand the tube on the supplied colour card and look down through the sample from directly above. Use indirect light and avoid direct sunlight. Keep the original card with the reagent so that each test is compared with the intended reference colours.
A sample can fall between two printed colours. Record the closest supported reading or note the interval when the match is uncertain. Do not claim extra precision simply because a written record has space for more decimal places. A photograph of the tube is not a substitute for making the comparison in the correct lighting and viewing position.
Sample volume, drop counting and clean equipment
The 5ml water volume and five reagent drops belong together in this method. Using the larger sample required for another kit while retaining the nitrite drop count changes the test conditions. Check the volume first, then hold the reagent bottle vertically while dispensing drops, as Colombo advises.
The manufacturer also calls for drops without air bubbles and a tube that is clean before use and cleaned immediately afterwards. If the count is interrupted or you cannot remember whether reagent has already been added, start again with fresh water. Guessing a correction can leave you with a result that is difficult to interpret reliably.
Linking the result to biological filtration
An ammonia test provides a useful companion reading because Colombo describes ammonia as the substance from which nitrite is produced during nitrification. The Colombo NH3 ammonia kit supplies that separate measurement. Recording both avoids assuming that one result answers every question about the filter’s performance.
Note the date, sample source and any recent changes such as filter maintenance or stocking. If seeking help with an unexpected result, those details are more useful than a number without context. Repeat an unexpected test carefully, while taking any signs of fish distress seriously rather than relying solely on how clear the water looks.
What the kit does and does not provide
This product measures nitrite; it does not remove it, establish the cause of every elevated reading or provide a treatment dose. Keep the actual reading and kit identity available when asking for pond-care advice. Do not assume a zero result for one parameter confirms every aspect of water quality.
